Overview

Haploidentical IL-15–activated expanded NK cells (NKAE cells) are an adoptive cell immunotherapy consisting of natural killer (NK) cells harvested from haploidentical donors. These cells are activated and expanded ex vivo using interleukin-15 (IL-15) and K562-mb15-41BBL feeder cells to enhance their cytotoxic potential and proliferation. The therapy leverages the graft-versus-leukemia effect, where donor NK cells target leukemic cells that lack matching self-MHC ligands (KIR-ligand mismatch). Developed by Antonio Pérez Martínez at Hospital Universitario La Paz, this treatment is primarily investigated for pediatric and young adult patients with relapsed or refractory acute leukemia, often in combination with salvage chemotherapy and post-infusion low-dose IL-2 to support cell persistence.
