Overview

This investigational adoptive cellular immunotherapy, developed by the M.D. Anderson Cancer Center, involves the infusion of natural killer (NK) cells derived from haploidentical (partially matched) related donors. The therapy is specifically being evaluated for patients with chronic myeloid leukemia (CML) who have failed to achieve cytogenetic remission after treatment with at least two tyrosine kinase inhibitors (TKIs), or those who have progressed to the accelerated or blast phases. The treatment protocol typically involves a lymphodepleting conditioning regimen (such as fludarabine and busulfan) followed by the infusion of donor NK cells and a subsequent allogeneic stem cell transplant. The donor NK cells are intended to provide a graft-versus-leukemia (GVL) effect by recognizing and eliminating residual malignant cells, often supported by the administration of Interleukin-2 (IL-2) to enhance cell survival and expansion.
