Overview

This investigational combination therapy, developed by the Masonic Cancer Center at the University of Minnesota, consists of allogeneic haploidentical natural killer (NK) cells, the IDO1 inhibitor epacadostat (INCB024360), and interleukin-2 (IL-2). It is specifically designed for the treatment of recurrent ovarian, fallopian tube, and primary peritoneal cancers. The regimen involves the intraperitoneal administration of donor-derived NK cells to provide direct anti-tumor activity within the peritoneal cavity. Epacadostat is administered orally to block the immunosuppressive effects of the IDO1 enzyme, which is often overexpressed in these tumors to inhibit immune responses. Interleukin-2 is co-administered intraperitoneally to enhance the survival, persistence, and cytotoxic function of the infused NK cells. This approach aims to overcome the immunosuppressive tumor microenvironment and leverage the innate anti-tumor capabilities of NK cells.
