Overview

Haploidentical NK cell therapy is an adoptive cellular immunotherapy developed by Seoul National University Hospital for the treatment of refractory or relapsed malignant melanoma. The therapy involves the infusion of ex vivo expanded natural killer (NK) cells derived from a haploidentical (partially HLA-matched) donor. This approach leverages the 'missing-self' hypothesis, where the mismatch between the donor's killer cell immunoglobulin-like receptors (KIRs) and the recipient's HLA ligands prevents the inhibition of NK cells, thereby enhancing their ability to recognize and lyse tumor cells. The cells are typically processed via CD3+ depletion to minimize the risk of graft-versus-host disease (GvHD) before expansion and infusion.
