HAT-CyAl5 is a hyaluronan-targeted near-infrared fluorescence (NIRF) molecular imaging probe designed for the detection of atherosclerotic plaque erosion (PE). It consists of a hyaluronan-binding peptide sequence conjugated to the NIR fluorophore CyAl5 (excitation/emission 643/661 nm). The agent specifically targets the enrichment of hyaluronic acid (HA) at the luminal interface of plaques, a pathological feature that distinguishes plaque erosion from other causes of acute coronary syndrome, such as plaque rupture. Developed by researchers at Massachusetts General Hospital and the Masonic Medical Research Institute, HAT-CyAl5 is intended for use with intravascular NIRF imaging to provide high-resolution visualization of plaque pathobiology in vivo. Preclinical studies in mouse models and human carotid specimens have demonstrated its ability to specifically bind HA and differentiate erosion-prone plaques.
