HB-06510 is a novel, fungal-derived natural product that acts as a potent protein translation inhibitor. Developed by Hexagon Bio using their proprietary machine learning and synthetic biology platform, HB-06510 is designed for use as a cytotoxic payload in antibody-drug conjugates (ADCs). It exhibits high potency against cancer cell lines, including multi-drug resistant multiple myeloma cells, with an average EC50 of ≤ 3.2 nM. The compound possesses favorable physicochemical properties for ADC development, such as good solubility, low lipophilicity, and stability in human plasma and lysosomal environments. Preclinical studies have demonstrated its efficacy when conjugated to antibodies targeting myeloma antigens such as BCMA, CD38, and CS1 (SLAMF7), inducing immunogenic cell death and tumor growth reduction.
