HB-06510 ADC is an antibody-drug conjugate (ADC) being developed by Hexagon Bio for the treatment of multiple myeloma. The drug consists of a monoclonal antibody targeting myeloma-associated antigens—specifically BCMA, CS1 (SLAMF7), or CD38—conjugated to HB-06510, a novel small molecule natural product payload. HB-06510 was identified through Hexagon Bio's proprietary platform that mines fungal natural products and acts as a potent protein translation inhibitor. Preclinical studies have shown that HB-06510 is highly cytotoxic (EC50 ≤ 3.2 nM) across various myeloma cell lines, including those resistant to standard therapies and those expressing efflux pumps like Pgp and ABCG2. The ADC has demonstrated significant anti-tumor activity and tumor growth reduction in both in vitro and in vivo multiple myeloma models.
