Overview

HB-110 is a **second-generation therapeutic DNA vaccine** in development for chronic hepatitis B infection. It is an **adenoviral-based DNA vaccine** that incorporates a gene fusion for **interleukin-12 (IL-12)** as an immunomodulatory adjuvant. The vaccine is administered via **electroporation**, which aims to increase membrane penetrability and enhance immune response. In clinical trials, HB-110 was combined with nucleos(t)ide analogues (such as adefovir dipivoxil or entecavir). HB-110 was designed to induce HBV-specific T-cell responses, especially CD4+ memory T-cell responses, and promote HBeAg seroconversion. Data have suggested that, while HB-110 is generally well tolerated, its immunogenicity may be lower in some populations due to higher levels of immune tolerance, such as in Korean patients with a high rate of vertical HBV transmission[2][1].
