HB0028 is a bispecific antibody designed as a bifunctional fusion protein that targets both programmed death-ligand 1 (PD-L1) and transforming growth factor beta (TGF-β). It consists of an anti-PD-L1 IgG1 single domain antibody fused to the extracellular domain of the TGF-β receptor II. The drug acts as both a PD-L1 inhibitor and a TGF-β inhibitor, aiming to enhance antitumor immune responses by blocking two key immunosuppressive pathways in the tumor microenvironment. Developed primarily for oncology indications, it is being investigated for use in advanced malignant solid neoplasms and uterine cervical cancer. Clinical studies have shown that HB0028 is generally well tolerated with modest antitumor activity in heavily pretreated patients with advanced solid tumors[1][2][5].
