HB0034 is a monoclonal antibody targeting the interleukin-36 receptor (IL-36R), developed by Shanghai Hile Bio-Pharmaceutical. It is currently in early-stage clinical development for the treatment of generalized pustular psoriasis (GPP), a rare and severe autoinflammatory skin disease characterized by the sudden eruption of sterile pustules. By binding to IL-36R, HB0034 acts as an antagonist, blocking the pro-inflammatory signaling of IL-36 cytokines (IL-36α, β, and γ) which are central to the pathogenesis of GPP. The drug is being evaluated in Phase 1 clinical trials to assess its safety, tolerability, pharmacokinetics, and immunogenicity in healthy subjects.
