HB0036 is a first-in-class bispecific monoclonal antibody designed to simultaneously target and inhibit two key immune checkpoints—programmed death-ligand 1 (PD-L1) and T cell immunoglobulin and ITIM domain (TIGIT). By dual blockade of the PD-L1/PD-1 and TIGIT/CD155 pathways, HB0036 aims to overcome tumor-induced immune suppression, thereby enhancing the activity of both T cells and natural killer (NK) cells. The molecule retains antibody-dependent cellular cytotoxicity (ADCC) functions for both targets, potentially enabling direct killing of tumor cells as well as regulatory T cells. Preclinical studies have shown that HB0036 has linear pharmacokinetics, a long half-life, favorable safety profile, and superior efficacy compared to monotherapies targeting either PD-L1 or TIGIT alone. It is being developed primarily for advanced solid tumors including non-small cell lung cancer (NSCLC), esophageal squamous cell carcinoma, among others[2][3][4][5][6].
