Overview

HB0043 is a bispecific monoclonal antibody developed to simultaneously inhibit the interleukin-17A (IL-17A) and interleukin-36 receptor (IL-36R) pathways. It was engineered by linking single chain fragment variables from a humanized anti-IL-36R antibody (HB0034) to the C-terminus of the heavy chain of an anti-IL-17A IgG1 antibody (HB0017), using a flexible peptide linker. This dual blockade approach aims to enhance anti-inflammatory activity beyond what is achievable with single cytokine inhibition, as IL-17 and IL-36 can cooperate to amplify pro-inflammatory and pro-fibrotic gene expression in skin cells. Preclinical studies demonstrated that HB0043 more effectively blocked inflammatory cytokine production compared to its parent antibodies, and animal models showed improved outcomes in skin inflammation without increased toxicity. The drug is being developed by Huabo Biopharm for immune-mediated inflammatory diseases, with clinical trials underway for hidradenitis suppurativa.
