Overview

HB1148 is a selective, small molecule inhibitor of ubiquitin-specific protease 7 (USP7), also known as HAUSP. USP7 is a deubiquitinating enzyme that regulates the stability of several key proteins, most notably MDM2, which in turn controls the levels of the tumor suppressor p53. By inhibiting USP7, HB1148 facilitates the ubiquitination and subsequent proteasomal degradation of MDM2, leading to the stabilization and activation of p53, which triggers cell cycle arrest and apoptosis in cancer cells. Beyond the p53 pathway, USP7 inhibition is thought to impact the stability of other oncogenic proteins and modulate the immune microenvironment, specifically by impairing the function of regulatory T cells (Tregs). Developed by Huabing Pharmaceutical, HB1148 is currently undergoing Phase 1 clinical evaluation for the treatment of advanced solid tumors.
