Overview

HB229 is a recombinant humanized monoclonal antibody developed by Hualan Biological Engineering as a biosimilar candidate for pertuzumab (Perjeta). It specifically targets the extracellular dimerization domain (Subdomain II) of the Human Epidermal Growth Factor Receptor 2 (HER2). By binding to this specific epitope, HB229 sterically hinders the heterodimerization of HER2 with other members of the HER family, most notably HER3. This inhibition prevents the activation of downstream signaling cascades, including the mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K) pathways, which are critical for the growth and survival of HER2-overexpressing cancer cells. HB229 is primarily being evaluated in Phase 3 clinical trials for the treatment of HER2-positive metastatic breast cancer, typically administered in combination with trastuzumab and docetaxel.
