Drug intelligence / Profile preview
HB4-6 is a first-generation humanized antagonist monoclonal antibody targeting CD84 (SLAMF5), a self-binding immunoreceptor belonging to the signaling lymphocyte activation molecule (SLAM) family. Developed through a collaboration between City of Hope and the Weizmann Institute of Science, and licensed to Slam BioTherapeutics, HB4-6 blocks CD84-mediated homophilic interactions. In multiple myeloma, CD84 is expressed on myeloma cells, immunosuppressive myeloid cells, and T cells, where it acts to suppress the immune response. By blocking CD84, HB4-6 reduces the expression of immune checkpoints (such as PD-L1, PD-1, and CTLA-4), increases T-cell and NK-cell activity, and induces macrophage-mediated programmed cell removal (PrCR) and antibody-dependent cellular cytotoxicity (ADCC) against tumor cells.
