**HBI-201** is an investigational oral liquid formulation of nitrous oxide (N2O) designed for safe, controlled delivery to treat acute pain associated with vaso-occlusive crises (VOC) in sickle cell disease (SCD). Developed by Hillhurst Biopharmaceuticals using their proprietary GLASS platform, it leverages the well-established analgesic properties of N2O—an FDA-designated medical gas—while avoiding risks of inhalation such as inadvertent exposure, toxicity, and addiction. Preclinical studies in mouse models of SCD demonstrate rapid, dose-dependent reversal of cold-induced mechanical and thermal hyperalgesia, spontaneous nociceptive behavior, and ongoing pain, with effects lasting ~90 minutes and peak blood N2O levels far lower than inhaled administration. Unlike opioids or inhaled N2O, HBI-201 shows no abuse liability (no conditioned place preference), no analgesic tolerance after 9 days of repeated dosing, and no sedation or motor impairment at effective doses (up to 20 mg/kg oral).[1][2][3]
