HBM1022 is an afucosylated human monoclonal antibody targeting C-C motif chemokine receptor 8 (CCR8), developed by Harbour BioMed. It is designed to selectively deplete tumor-infiltrating regulatory T cells (Tregs) that express CCR8 via antibody-dependent cell-mediated cytotoxicity (ADCC), thereby enhancing anti-tumor immune responses. Preclinical studies have demonstrated potent antitumor activity both as monotherapy and in combination with anti-PD-(L)1 antibodies such as pembrolizumab. The drug has shown favorable pharmacokinetic properties and safety profiles in animal models, including cynomolgus monkeys. Its primary indication is for the treatment of solid tumors enriched in CCR8-positive Tregs, with potential applications across various cancer types including breast cancer, colon cancer, gastric cancer, non-small cell lung cancer, head and neck cancers, bladder cancer, renal cell carcinoma and other solid tumors[1][3][4][5][6].
