HBM7004 is a novel bispecific antibody developed using the HBICE (HCAb Based Immune Cell Engagers) platform by Harbour BioMed. It targets B7-H4, a tumor-associated antigen overexpressed in various solid tumors, and CD3 on T cells. The molecule features a unique "2+1" asymmetric format with two B7H4 binding moieties and one optimized CD3 binding moiety. This design enables selective engagement of T cells to B7H4-positive tumor cells, promoting targeted T-cell dependent cellular cytotoxicity (TDCC) while minimizing systemic toxicity due to weak CD3 affinity and avidity-dependent B7H4 binding. Preclinical studies have demonstrated strong anti-tumor efficacy, remarkable in vivo stability, reduced systemic toxicity, and synergistic effects when combined with other immune cell engagers such as B7H4x4-1BB bispecific antibodies[2][5][6]. The drug is being developed for the treatment of solid tumors.
