HBM7022 is a fully human bispecific antibody designed to crosslink tumor cells and T cells by simultaneously targeting Claudin 18.2 (CLDN18.2), a tumor-associated antigen highly expressed in certain cancers, and CD3 on T cells. This dual targeting leads to potent T cell activation and subsequent tumor cell elimination. The molecule uses bivalent high-affinity anti-Claudin 18.2 arms and a monovalent low-affinity anti-CD3 arm to maximize cytotoxicity while minimizing the risk of cytokine release syndrome[4][6]. Developed using Harbour BioMed’s HBICE platform, HBM7022 has demonstrated preclinical efficacy against wild-type Claudin 18.2-positive gastric cancer, mutated Claudin 18.2 gastric cancer, and pancreatic cancer[4][6]. It is being developed for advanced solid tumors with an initial focus on gastric carcinoma and esophageal adenocarcinoma[1][4].
