HBM9027 is a novel, fully human bispecific antibody that targets both programmed death-ligand 1 (PD-L1) and CD40. Developed using Harbour BioMed’s proprietary HBICE® bispecific technology and Harbour Mice® platform, it is designed to activate the CD40 pathway only in the presence of PD-L1 crosslinking. This mechanism aims to provide potent immune modulation with improved safety by restricting CD40 activation to tumor sites where PD-L1 is overexpressed, thereby reducing systemic toxicity commonly associated with traditional anti-CD40 monoclonal antibodies. Preclinical studies have demonstrated that HBM9027 exhibits crosslinking-dependent specificity for tumors, robust immune modulation activity, strong antitumor efficacy in vivo, and a favorable safety profile. The drug mediates both inhibition of the PD-1/PD-L1 pathway and agonism of the CD40 pathway for synergistic antitumor responses. It is being developed primarily for advanced solid tumors[1][2][3][4][5][6][8].
