HBM9033 is an antibody-drug conjugate (ADC) that specifically targets human mesothelin (MSLN), a tumor-associated antigen upregulated in various solid tumors, including mesothelioma, ovarian cancer, lung cancer, breast cancer, and pancreatic cancers. The drug consists of a fully human monoclonal antibody generated using the Harbour Mice platform that preferentially binds to membrane-bound MSLN over soluble forms. This design minimizes interference from shed antigen and enhances tumor targeting. The ADC employs a tumor-specific cleavable linker attached to a novel topoisomerase inhibitor payload for improved stability and anti-tumor activity. Preclinical studies have demonstrated superior potency and safety compared to other agents in this class. HBM9033 was developed by Harbour BioMed in collaboration with MediLink Therapeutics; Nona Biosciences has also been involved as an originator/developer, with Pfizer holding global license rights for further development[1][5][6][7].
