Overview
GSK2231392A is an investigational prophylactic vaccine developed by GSK for the prevention of hepatitis B virus (HBV) infection. It consists of recombinant hepatitis B surface antigen (HBsAg) formulated with GSK's proprietary AS01B Adjuvant System. The AS01B adjuvant is a liposome-based system containing two immunostimulants: 3-O-desacyl-4'-monophosphoryl lipid A (MPL), a TLR4 agonist, and QS-21, a saponin extracted from *Quillaja saponaria*. This combination is designed to enhance both humoral and cell-mediated immune responses against the HBV surface antigen. The vaccine has been evaluated in Phase II clinical trials (e.g., NCT01777295) to assess its safety, reactogenicity, and immunogenicity compared to standard aluminum-adjuvanted vaccines like Engerix-B.
