HBW-012336 is an orally bioavailable small molecule inhibitor specifically targeting the KRAS G12D mutation, developed by Chengdu Hyperway Pharmaceutical. KRAS G12D is a prevalent driver mutation in several difficult-to-treat malignancies, including pancreatic, colorectal, and non-small cell lung cancers. HBW-012336 is designed for high potency and selectivity, demonstrating superior preclinical anti-tumor efficacy compared to the competitor compound MRTX1133. In mouse xenograft models, it has shown the capacity to completely inhibit tumor growth or reduce tumor volume, and it exhibits synergistic effects when combined with cetuximab in colorectal cancer models. The drug entered Phase I clinical trials in 2025 following approval by the NMPA.
