HC-067047 is a potent and selective small molecule antagonist of the Transient Receptor Potential Vanilloid 4 (TRPV4) receptor. It inhibits TRPV4-mediated calcium influx with nanomolar potency across human, rat, and mouse orthologs. In preclinical research, HC-067047 has demonstrated therapeutic potential in animal models of bladder overactivity, cyclophosphamide-induced cystitis, and chronic cough. Additionally, recent studies suggest it may suppress the metastasis of hepatocellular carcinoma by remodeling single-cell polarity. Although widely utilized as a pharmacological tool to study TRPV4 function, it has not progressed to clinical development for human use and is primarily supplied as a research reagent.
