HC029-CPT is a novel, conformation-selective antibody-drug conjugate (ADC) targeting Integrin beta-6 (ITGB6), developed by Hongcheng Biopharma. It consists of HC029, an activation-state-selective anti-ITGB6 monoclonal antibody (Fc-silenced human IgG1), conjugated to camptothecin (CPT) derivatives via an extra-stable linker. Unlike conventional ITGB6-targeted therapies, HC029-CPT specifically binds to a unique conformational epitope exposed on activated ITGB6 in the tumor microenvironment, achieving high selectivity for disease-specific states while sparing physiological ITGB6. Preclinical studies in non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), and bladder cancer (BLCA) models have demonstrated potent antitumor activity. Toxicology studies in non-human primates indicated a favorable safety profile with a maximum tolerated dose (MTD) exceeding 60 mg/kg and minimal systemic payload release.
