HC031 is an antibody-drug conjugate (ADC) targeting A Disintegrin And Metalloproteinase 9 (ADAM9), a transmembrane protein overexpressed in various solid tumors and linked to tumor progression and poor prognosis. Developed by Hengrui Pharma, HC031 consists of an affinity-optimized, humanized monoclonal antibody conjugated to a proprietary camptothecin derivative payload via a highly stable linker. The antibody component is designed to selectively bind the membrane-bound form of ADAM9 while avoiding the soluble form, which helps maintain a regular pharmacokinetic profile. Additionally, its binding is partially dependent on bivalent cations, potentially enhancing its safety profile. Preclinical studies have demonstrated that HC031 induces robust tumor regression in xenograft models and possesses a high maximum tolerated dose (MTD) of 120 mg/kg in non-human primates, indicating a broad therapeutic window.
