Overview

HCB301 is a tri-specific recombinant fusion protein designed for cancer immunotherapy. It consists of an engineered IgV domain of human SIRPα fused to an anti-PD-L1 (Ig) V domain and an engineered human TGFβ receptor II (TGFRII). This structure enables simultaneous blockade of the CD47/SIRPα axis and PD-L1 checkpoint while also trapping and inhibiting TGFβ-mediated immune suppression. The goal is to enhance anti-tumor immune responses by overcoming resistance mechanisms associated with current checkpoint inhibitors. Preclinical studies have shown favorable safety profiles, particularly regarding reduced risk of anemia or cytopenias due to selective tumor cell binding over red blood cells. The drug is being developed primarily for advanced solid tumors and relapsed/refractory classical Hodgkin lymphoma[1][3][4][5].
