Overview

hCD19 CAR-T is an autologous, humanized chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen. Developed by the Affiliated Hospital of Xuzhou Medical University, it is primarily investigated for the treatment of hematologic malignancies, often in combination with other CAR-T therapies such as those targeting BCMA. The 'h' in hCD19 signifies the use of a humanized single-chain variable fragment (scFv) in the CAR construct, which is designed to minimize the host immune response against the CAR-T cells, potentially leading to improved persistence and reduced risk of rejection compared to murine-based CARs. In clinical settings, it has been studied as a consolidation therapy for multiple myeloma patients who have not achieved complete remission following induction chemotherapy, frequently administered alongside or in comparison to autologous hematopoietic stem cell transplantation (ASCT).
