HCI-1401 is an orally bioavailable, small-molecule, irreversible inhibitor of Bruton's tyrosine kinase (BTK) being developed primarily for the treatment of B‑cell malignancies and potentially autoimmune diseases. It was discovered at Huntsman Cancer Institute and exclusively licensed to LSK BioPharma (now Elevar Therapeutics) for further development and commercialization worldwide.[1][7][8][10] Preclinical in vitro data indicate that HCI-1401 is several-fold more potent against BTK than first-generation BTK inhibitors, with the goal of providing deeper and more durable B‑cell pathway inhibition while retaining a favorable safety profile for chronic administration.[1][8][10]
