HCR-688

HCR-688 is an antibody-peptide conjugate (APC) being developed by Helicore Biopharma for the treatment of obesity and cardiometabolic diseases. It is built upon the HCR-188 monoclonal antibody backbone, which functions as a ligand binder to deplete circulating glucose-dependent insulinotropic polypeptide (GIP). This antibody is conjugated to a selective amylin receptor type 1 (AMY1R) agonist payload. The AMY1R agonist is highly specific, with reported selectivity of greater than 10,000-fold over AMY3R and negligible activity at the calcitonin gene-related peptide (CGRP) receptor. This dual-mechanism approach is designed to provide potent weight loss and metabolic improvement with a long-acting pharmacokinetic profile that may support quarterly or less frequent dosing.
