**HCW11-018b** is a second-generation tetra-valent bispecific T-cell engager (BiTE) developed by HCW Biologics using their proprietary TRBC platform. It comprises anti-tissue factor (TF)/anti-CD3 single chain antibody domains for targeted T-cell activation at tumor sites, soluble TGFβRII (TGF-β trap) to neutralize immunosuppressive TGF-β signaling in the tumor microenvironment, and soluble IL-15/IL-15RαSu complex to expand and activate exhausted tumor-infiltrating T cells, including progenitor exhausted T (TPEX) cells. Preclinical studies in pancreatic cancer xenograft and patient-derived xenograft (PDX) models demonstrated 100% survival in treated tumor-bearing mice (versus 0% in untreated), tumor shrinkage, enhanced immune cell infiltration and cytotoxicity, broad applicability to solid tumors, favorable pharmacokinetics with long half-life, subcutaneous tolerability without cytokine storm, and streamlined manufacturing similar to monoclonal antibodies.[1][4][5][7][8][9]
