HCW11-040 is an investigational, heterodimeric tetrafunctional recombinant fusion immunotherapeutic developed by HCW Biologics using its soluble human T-cell receptor beta constant domain scaffold. The single molecular entity combines a pembrolizumab-derived anti-programmed cell death protein 1 diabody, interleukin-7, an interleukin-15/interleukin-15 receptor alpha complex, and extracellular transforming growth factor beta receptor type II domains that function as a transforming growth factor beta trap. It is designed to block programmed cell death protein 1/programmed death-ligand 1 signaling, stimulate T cells and natural killer cells through cytokine signaling, and relieve transforming growth factor beta-mediated immunosuppression in the tumor microenvironment.
