Drug intelligence / Profile preview
HCXT-2001 + emvistegrast is a proposed all-oral combination regimen under investigation for the treatment of inflammatory bowel disease (IBD), including ulcerative colitis and Crohn's disease. The combination pairs HCXT-2001, an oral macrocyclic peptide inhibitor of the interleukin-23 receptor (IL-23R) originally developed by PepStar Therapeutics, with emvistegrast (GS-1427), Gilead Sciences' internally developed oral small-molecule inhibitor of the α4β7 integrin. By combining an IL-23R blocker with an integrin inhibitor, the regimen aims to target two distinct, clinically validated pathways in IBD to achieve synergistic therapeutic efficacy. Gilead acquired the oral IL-23R program from PepStar in October 2026 and has proposed the combination as a potential once-daily oral regimen, though clinical testing of the combination has not yet commenced.
