Drug intelligence / Profile preview
HCXT-2001 + GS-1427 is an investigational oral combination therapy being developed by Gilead Sciences for the treatment of inflammatory bowel disease (IBD), including ulcerative colitis and Crohn's disease. The combination consists of HCXT-2001, an oral macrocyclic peptide inhibitor of the interleukin-23 receptor (IL-23R) originally developed by PepStar Therapeutics, and emvistegrast (GS-1427), an oral small molecule inhibitor of the α4β7 integrin developed internally by Gilead. By simultaneously targeting the IL-23 pathway (which drives chronic mucosal inflammation) and the α4β7 integrin pathway (which mediates the trafficking of T cells to inflamed gut tissues), this dual-action oral regimen aims to provide deeper and more durable clinical remission in patients with moderate-to-severe IBD without systemic immunosuppression.
