HD-10019

**HD-10019** is a rationally designed **type II kinase inhibitor** developed for **FLT3-mutated acute myeloid leukemia (AML)**. It targets resistance mutations in the **FLT3 tyrosine kinase domain**, offering improved selectivity over standard-of-care agents like gilteritinib to reduce myelosuppression while avoiding cardiac toxicities associated with other FLT3 inhibitors. Licensed by **Harmonic Discovery** from **BioVentures** (University of Arkansas for Medical Sciences) and **UC San Francisco**, it has shown promising in vivo efficacy and a potentially best-in-class profile in preclinical studies, with IND-enabling studies planned for late 2024.[1][2][3][11]
