Drug intelligence / Profile preview
HDAC6 inhibitor #60 is a selective histone deacetylase 6 (HDAC6) inhibitor featuring a quinazolinone-based structure and a difluoromethyl oxadiazole zinc-binding group. Developed by researchers at National Taiwan University and MacKay Memorial Hospital, the compound is designed to selectively target HDAC6 while reducing the side effects typically associated with pan-HDAC inhibitors. In preclinical models of chronic kidney disease (CKD), specifically unilateral ureteral obstruction (UUO)-induced renal fibrosis, oral administration of HDAC6 inhibitor #60 has been shown to restore acetylation of histone H3 and alpha-tubulin, downregulate fibrotic markers (such as alpha-SMA, PDGFR-beta, collagen I, collagen III, and TGF-beta1), and significantly attenuate renal fibrosis.
