Overview

HDAC6 shRNA (short hairpin RNA) is a genetic tool designed to silence the expression of the Histone Deacetylase 6 (HDAC6) gene through RNA interference. In preclinical oncology research, particularly in melanoma, HDAC6 shRNA is utilized to investigate the role of this enzyme in tumor progression, cell cycle regulation, and immune signaling. By knocking down HDAC6 protein levels, researchers have observed inhibited cell proliferation, G1 phase cell cycle arrest, and altered phosphorylation and acetylation of STAT1 and STAT3 proteins within the JAK/STAT pathway. Studies conducted at the H. Lee Moffitt Cancer Center & Research Institute have demonstrated that melanoma cells treated with HDAC6 shRNA exhibit reduced growth in both in vitro cell lines and in vivo murine models, identifying HDAC6 as a potential therapeutic target.
