Overview

HDL-MDP nanoparticles are an early-phase nanobiologic research compound consisting of high-density lipoprotein (HDL)-mimetic nanoparticles loaded with muramyl dipeptide (MDP), a bacterial cell wall peptidoglycan component and well-characterized agonist of trained immunity. MDP signals through the intracellular pattern recognition receptor NOD2 (nucleotide-binding oligomerization domain-containing protein 2), inducing epigenetic and metabolic reprogramming of innate immune cells such as monocytes and myeloid progenitor cells. In a Dutch translational clinical study (NCT05280379) conducted by Radboud University Medical Center, HDL-MDP nanoparticles were evaluated as an ex vivo trained-immunity-inducing agent to assess whether circulating monocytes and bone marrow myeloid progenitor cells from adults with newly diagnosed thyroid carcinoma, colon carcinoma, or melanoma could be transcriptionally, epigenetically, and functionally reprogrammed. The study was prematurely terminated, and the compound remains a preclinical research tool.
