Overview

HDM-SPIRE is a synthetic peptide-based intradermal immunotherapy developed for the treatment of house dust mite-induced allergic rhinoconjunctivitis. It consists of short peptides derived from major house dust mite allergens, designed to induce immune tolerance by modulating T-cell responses. The mechanism involves binding to MHC class II molecules on antigen-presenting cells, leading to upregulation of regulatory T-cells and suppression of IgE-mediated allergic responses. This approach aims to reduce the risk of IgE-mediated side effects compared with traditional allergen immunotherapies and potentially offers a shorter course of treatment. The drug was originally developed by Circassia (now NIOX), but development for allergic rhinoconjunctivitis has been discontinued after phase 2 trials[1][6][8].
