Overview

HDM1014 is a GalNAc-conjugated small interfering RNA (siRNA) designed to target and knock down the expression of Inhibin-β E (INHBE) in the liver. Developed by Huadong Medicine, it is intended for weight management and the treatment of metabolic disorders. INHBE is a member of the TGF-β superfamily that suppresses lipolysis; its inhibition is associated with fat-selective weight loss and the preservation of lean muscle mass, addressing a common limitation of GLP-1 receptor agonists. Preclinical studies in non-human primates and mice have demonstrated robust and durable hepatic INHBE mRNA knockdown, leading to significant reductions in body fat without muscle wasting. HDM1014 has also shown synergistic effects when combined with GLP-1/GIP receptor agonists like HDM1005.
