HDM2012 is an oral small molecule inhibitor of Hematopoietic Progenitor Kinase 1 (HPK1), currently under development by Huadong Medicine for the treatment of advanced solid tumors. HPK1 is a member of the MAP4K family and acts as a negative regulator of T-cell receptor (TCR) signaling. By inhibiting HPK1, HDM2012 aims to enhance T-cell activation and anti-tumor immune responses, potentially overcoming immunosuppression in the tumor microenvironment. It is being evaluated as a monotherapy and potentially in combination with other immunotherapies in Phase I clinical trials.
