Overview

HDM3706 is a recombinant fusion protein developed by Huadong Medicine for the treatment of IgA nephropathy (IgAN). It functions as a dual-target inhibitor, simultaneously neutralizing B-cell activating factor (BAFF) and a proliferation-inducing ligand (APRIL). These two cytokines are essential for the survival, proliferation, and differentiation of B cells and plasma cells. In IgAN, the overproduction of galactose-deficient IgA1 (Gd-IgA1) by these cells leads to the formation of immune complexes that deposit in the glomerular mesangium, causing renal inflammation and progressive kidney damage. By inhibiting both BAFF and APRIL, HDM3706 aims to more effectively reduce the levels of pathogenic IgA1 and provide a therapeutic benefit over single-target therapies. The drug is currently in Phase 1 clinical trials following IND approval in China in early 2024.
