HDP-101 is a novel antibody-drug conjugate (ADC) designed to target B-cell maturation antigen (BCMA) on multiple myeloma cells. It consists of an anti-BCMA monoclonal antibody linked to a synthetic amanitin payload, which acts as a potent inhibitor of RNA polymerase II. By inhibiting RNA polymerase II, the drug halts transcription and induces apoptosis in tumor cells, including those that are non-proliferating or have low BCMA density. Preclinical studies have shown that HDP-101 is effective against both proliferating and dormant myeloma cells and can overcome drug resistance. The agent has demonstrated selective cytotoxicity for BCMA-positive myeloma cells while sparing BCMA-negative cells, with promising results in animal models and early-phase clinical trials[1][2][4][5][8].
