HDP-201

HDP-201 is an antibody-drug conjugate (ADC) developed by Heidelberg Pharma. It utilizes exatecan, a DNA topoisomerase I inhibitor, as its cytotoxic payload and targets guanylyl cyclase-C (GCC), a receptor highly expressed on the surface of intestinal cells and in various gastrointestinal cancers—including over 95% of colorectal cancers and significant proportions of esophageal, gastric, and pancreatic tumors. The ADC employs a multimeric linker technology to deliver exatecan directly to GCC-expressing tumor cells. Preclinical studies have demonstrated that HDP-201 exhibits strong target-specific potency, complete tumor remission in GCC-expressing models, significant inhibition of tumor growth in patient-derived xenograft models for gastrointestinal cancer, and high tolerability at tested doses. The drug is currently being developed primarily for colorectal cancer[1][3][4][5][7][9].
