Overview

HE3177 is a synthetic small molecule androstene derivative developed by Hollis-Eden Pharmaceuticals (later Harbor BioSciences) as part of their immune-regulating hormone (IRH) platform. Designed as a second-generation analog of dehydroepiandrosterone (DHEA) metabolites, HE3177 was primarily investigated for the treatment of inflammatory conditions, most notably rheumatoid arthritis. The drug functions by modulating the production of key pro-inflammatory cytokines, such as tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6), without the androgenic or estrogenic side effects typically associated with steroid therapies. Although it entered Phase 1 clinical testing to evaluate safety and pharmacokinetics, development was ultimately discontinued following the developer's shift in strategic focus toward other IRH candidates and subsequent financial restructuring.
