HE3413 is a synthetic androstene steroid derivative developed by Hollis-Eden Pharmaceuticals (later Harbor BioSciences) as a potential treatment for castration-resistant prostate cancer (CRPC). Chemically identified as 17α-ethynyl-5-androstene-3β,7β,17β-triol, it is a 17α-ethynyl analog of a naturally occurring dehydroepiandrosterone (DHEA) metabolite. The compound was designed to address the limitations of traditional androgen deprivation therapy by inhibiting the intra-tumoral conversion of adrenal steroid precursors into potent androgens and by directly antagonizing the androgen receptor. Preclinical studies indicated that HE3413 could suppress tumor growth in hormone-refractory models and possessed anti-inflammatory properties. However, clinical development was discontinued following the financial decline and eventual dissolution of Harbor BioSciences.
