Overview

Heat-inactivated Escherichia coli is a microbial therapy being investigated for its potential to modulate immune responses, particularly in the context of breast cancer. It functions by stimulating macrophages, polarizing them towards a tumoricidal M1 phenotype. This immune modulation is mediated through bacterial components, such as lipopolysaccharide (LPS), which are known to interact with Toll-like receptors, specifically Toll-like receptor 4 and potentially Toll-like receptor 2. Preclinical studies have shown that macrophages stimulated with heat-inactivated E. coli can reduce tumor cell numbers in co-culture with breast cancer cell lines, suggesting a promising strategy for enhancing immunotherapy efficacy.
