Heat shock protein 70 small interfering RNA (HSP70 siRNA) is an RNA-based therapeutic approach designed to silence the expression of the HSPA1A gene, which encodes the major stress-inducible chaperone protein HSP70. HSP70 is frequently overexpressed in various malignancies, including hepatocellular carcinoma (HCC), where it facilitates protein folding, prevents apoptosis, and promotes tumor cell proliferation, migration, and metastasis. By utilizing the RNA interference (RNAi) pathway, the siRNA mediates the sequence-specific degradation of HSP70 mRNA. This downregulation leads to cell cycle arrest at the G1/S transition—often associated with the upregulation of p27Kip1 and downregulation of cyclin D1—thereby inhibiting the growth and invasive potential of cancer cells. While primarily used as a research tool to validate HSP70 as a therapeutic target, siRNA-mediated knockdown of HSP70 represents a potential strategy for precision oncology.
