Overview

The heat-treated zoster vaccine (V212) is an investigational inactivated viral vaccine developed by Merck for the prevention of herpes zoster (shingles) and its complications in immunocompromised individuals, such as recipients of autologous hematopoietic stem cell transplants (auto-HSCT) and patients with solid tumors undergoing chemotherapy. Based on the Oka/Merck strain of the varicella-zoster virus (VZV), the vaccine is inactivated by heat treatment to render it non-replicating, making it a safer alternative to the live-attenuated vaccine (Zostavax) which is contraindicated in immunocompromised patients. Although V212 demonstrated efficacy and safety in Phase III clinical trials, Merck discontinued its development in 2019 due to changes in the herpes zoster vaccine landscape, primarily driven by the approval and widespread adoption of GSK's highly effective recombinant adjuvanted vaccine, Shingrix.
