Drug intelligence / Profile preview
HEBE2 is a novel chimeric fusion protein (chronokine) designed to target multiple pathological pathways in Alzheimer's disease (AD) and other age-related neurodegenerative disorders. It is engineered by merging three soluble proteins: soluble Triggering Receptor Expressed on Myeloid Cells 2 (sTREM2), soluble Klotho (sKlotho or sKL), and TIMP metallopeptidase inhibitor 2 (TIMP2). Developed by researchers at the Autonomous University of Barcelona (UAB) and the Vall d'Hebron Research Institute (VHIR), HEBE2 is designed to overcome the limitations of single-target therapies. In preclinical models, HEBE2 has been shown to modulate microglial activation, enhance the phagocytosis of beta-amyloid monomers by ApoE4/4 microglia, increase the secretion of beneficial cytokines (such as IL-6, IL-12 p70, and IL-1b), extend health span, and partially restore altered molecular brain composition.
